Manuscript: Intra-articular injection of autologous tolerogenic dendritic cells modifies the synovial immune landscape in rheumatoid and inflammatory arthritis

Data Repository: Tolerogenic Dendritic Cell Effects on Synovial Immune Landscape

This collection contains data associated with the study investigating how intra-articular tolerogenic dendritic cells (tolDC) affect the local synovial immune landscape in autoimmune arthritis. This work builds upon our previously reported trial where high doses of tolDC appeared to stabilize knee symptoms, and specifically examines the synovial tissue changes using multi-parametric imaging mass cytometry (IMC).

The study analyzed paired synovial biopsies from 7 participants (5 tolDC treated and 2 control) taken at baseline and 14 days after intra-articular injection. Using a 27-antibody IMC panel defining lymphoid, myeloid, and stromal cells, we identified a significant tolDC-induced increase in synovial myeloid cells expressing high levels of the inflammation resolution marker Mer tyrosine kinase (MerTK-High), which correlated with tolDC dose and inversely correlated with arthroscopic synovitis scores.
